Determination of Crude Saponin By UV

Meliorate 4-Aminobenzaldehyde, Methanol, UV-VIS Spectrophotometer.

2. Standard preparation.

Accurately weight 0.6 of CoCl₂·6H₂0 to a 25ml flask. Dissolve with water and dilute to the volume.

3. Sample preparation:

Accurately weight 200mg of extract powder to a 10ml volumetric flask, Sonicate about 30min. Cool down and dilute to the volume. Filter and transfer 2ml filter to a 10ml volumetric flask. Add 3ml of methanol, 5ml of Meliorate 4-Aminobenzaldehyde. Then make a water bath about 2 hours. Cool to room temperature.

4. Blank preperation:

Transfer 5ml of methanol to a 10ml volumetric flask . Add 5ml of Meliorate 4-Aminobenzaldehyde. Then make a water bath about 2 hours. Cool to room temperature.

5.Analysis:

Turn on the spectrophotometer and adjust the wavelength to 512nm. Allow the instrument to warm up for thirty minutes.

Zero the instrument with blank solution. Read and record the absorbances.

  Content of saponins=(A_sample×W _CoCl2·6H2O)/( A_CoCl2·6H2O×2×W_sample) ×100%
